Dynamic modeling of folliculogenesis signaling pathways in the presence of miRNAs expression

چکیده
BACKGROUND TEK signaling plays a very important role in folliculogenesis. It activates Ras/ERK/MYC, PI3K/AKT/mTORC1 and ovarian steroidogenesis activation pathways. These are the main pathways for cell growth, differentiation, migration, adhesion, proliferation, survival and protein synthesis. RESULTS TEK signaling on each of the two important pathways where levels of pERK, pMYC, pAkt, pMCL1 and pEIF4EBP1 are increased in dominant follicles and pMYC is decreased in dominant follicles. Over activation of ERK and MYC which are the main cell growth and proliferation and over activation of Akt, MCl1, mTORC1 and EIF4EBP1 which are the main cell survival and protein synthesis factors act as promoting factors for folliculogenesis. In case of over expression of hsa-miR-30d-3p and hsa-miR-451a, MYC activity level is considerably increased in subordinate follicles. Our simulation results show that in the presence of has-miR-548v and bta-miR-22-3p, downstream factors of pathways are inhibited. CONCLUSIONS Our work offers insight into the design of natural biological procedures and makes predictions that can guide further experimental studies on folliculogenesis pathways. Moreover, it defines a simple signal processing unit that may be useful for engineering synthetic biology and genes circuits to carry out cell-based computation.

AKT family and miRNAs expression in IL-2-induced CD4+T cells
Objective(s): Study of non-coding RNAs is considerable to elucidate principal biological questions or design new therapeutic strategies. miRNAs are a group of non-coding RNAs that their functions in PI3K/AKT signaling and apoptosis pathways after T cell activation is not entirely clear. Herein, miRNAs expression and their putative targets in the mentioned pathways were studied in the activated ...
متن کاملThe Role of miRNA Dysregulation in Thyroid Cancer Development by Targeting the Main Signaling Pathways
Thyroid cancer is one of the most common malignancies of endocrine glands, causing carcinomas, such as papillary, follicular, medullary, and anaplastic thyroid carcinomas. Due to the significance of thyroid carcinomas, identification of the main signaling pathways and the affecting mutations has been considered by researchers. Further studies on the dysregulation of oncogenes in signaling path...
متن کاملPathway Analysis of miRNA-1 and Its Expres-sion Evaluation in Donor’s Serum from HIV-Positive Individuals vs Unaffected Controls
Background MicroRNAs (miRNAs) are non-coding RNA molecules (19-24 nucleotides) that play a major role in a wide range of biological processes through post-transcriptional regulation of gene expression. Differential expression of miRNAs has been reported in various infectious diseases such as HIV infection. The characterization of miRNA expression profiles, especially in mammalian biofluids, whi...
متن کاملLAT-derived microRNAs in HSV-1 target SMAD3 and SMAD4 in TGF-β/Smad signaling pathway
Background: During its latent infection, HSV-1 produces only a miRNA precursor called LAT, which encodes six distinct miRNAs. Recent studies have suggested that some of these miRNAs could target cellular mRNAs. One of the key cell signaling pathways that can be affected by HSV-1 is the TGF-β/Smad pathway. Herein, we investigated the potential role of the LAT as well as three LAT-derived miRNAs ...
متن کاملPrediction of MicroRNAs bind to Toll-like Receptors Pathway in Chicken based on Bioinformatics Method
Background: Toll-like receptors (TLRs) detect diverse pathogen-associated molecular patterns and play a critical role in the innate immune response. Hosts should activate TLR-signaling pathways to eliminate invading pathogens. However, excessive activation of these pathways may interrupt immune homeostasis, leading to several diseases. Therefore precise regulation of TLR-signaling pathways is e...
